Because man is born in sin since Adam, he could not come in contact with God (the ark represented God on earth) until the advent of Christ. 

In the same way the woman was not allowed to touch Jesus at the tomb until He ascended. She was unclean and Jesus had not yet been glorified until He returned to heaven. 

Now, we can speak to God/Jesus directly because He made the way for us to do so. He rent the curtain between us and God so that we can approach Him and He loves us when we do and are obedient.

As S. Michael Houdmann has already said, God had given particular instructions through Moses how the tabernacle was to be erected, dismantled, and moved (Numbers 4), and the main pieces of furniture were to be carried on the shoulders of the Levites who were descended from Kohath (Num. 4:9-20). However, when Uzza, David, and company used a new cart drawn by oxen, they were following the pattern of the pagan Philistines (1 Sam. 6), not the pattern given to Moses on Mount Sinai.

Doing God’s work God’s way brings God’s blessing. God’s work must be done in God’s way or there will be consequences. God had warned about taking hold of the ark to steady it (or at least about touching it) in the law of Moses, and every Israelite knew it (Num. 1:51; 4:15, 20).

Michael and the others did a great job of explaining the need to follow God's instructions. I think it goes a little deeper than that and simpler at the same time.

God is God! When he institutes a law the law is irrevocable. If this is not the case, then Jesus died for no reason. God issued a law that if you touched the ark you would die. If he had not died, God would not be God! God also said, "The wages of sin is death"! If Jesus had not fulfilled that law, we would all be sentenced to permanent death.

Jesus said that not one iota of the law would pass away until all is fulfilled. Jesus fulfilled the law by living without sin! Through Jesus and through Jesus alone we can be saved from the law!

Would mankind turn to God if he did not enforce His law? Would He even be God?

God struck Uzzah dead for touching the Ark of the Covenant because it was a direct violation of divine law, which strictly forbade touching the holy object upon penalty of death, regardless of good intentions. Uzzah's action demonstrated a lack of reverence for God's holiness. It resulted from transporting the Ark on a cart, in violation of the command that Levites must carry it on their shoulders. 

Some reasons for Uzzah's death based on the biblical account include:

Direct Disobedience of Law: Numbers 4:15 explicitly warned that the Kohathites (a Levite branch) must not touch the holy things directly, or they would die.

Irreverence for Holiness: Uzzah treated the presence of God casually, disregarding the strict separation between the holy Ark and human contact.

Improper Transportation Method: The Ark was being transported on a new cart pulled by oxen, whereas God commanded it to be carried on poles by Levites.

A Lesson on Compliance: The incident taught Israel that God's instructions require strict obedience rather than human innovation or "good intentions". 

While Uzzah's intention seemed to be protecting the Ark from falling, his action signalled a failure to trust in God's power to protect His own symbol.

A Lesson in Accountability

Uzzah's death served as a stark reminder to the nation of Israel that God's commands are to be followed precisely. 

Familiarity: Some suggest that because the Ark had stayed in Uzzah's family home (the house of Abinadab) for 20 years, he may have become too familiar with it, losing the appropriate "holy fear" and reverence required for such a sacred object.

Leadership Correction: After the incident, David was "afraid of the Lord" and eventually corrected the transport method, ensuring the Levites carried the Ark on their shoulders as originally commanded.

“Then the anger of the LORD was aroused against Uzzah, and He struck him because he put his hand to the ark; and he died there before God” (1 Chronicles 13:10).
