The phrase "born again" was used by Jesus in the passage from the gospel of John cited in the question while talking with Nicodemus, who was a member of the class of Jewish clerics known as Pharisees. Nicodemus thought that Jesus was referring to the process of physical human birth, and could not understand how someone could re-enter their mother's womb and be born again.

Jesus explained that the "new birth" that He was referring to was a spiritual process by which people came to realize that they had an inborn sin nature that estranged them from a holy God, and that had to be replaced with a new nature that would result in them having their sins (that is, their disobedience to God's commandments) forgiven through faith in Jesus (the Son of God who sacrificed His own life on their behalf to make that forgiveness possible), and then being indwelt by God the Holy Spirit. This process would put them in a right spiritual relationship with God, and would allow them to live eternally in His presence after their temporal lives on earth had ended in death.

It is with reference to that eternal life that is theirs through faith in Christ to which Christians are referring when they say that they have been "born again".

3. Another feeling of direction throughout everyday life.

"...just as Christ was raised from the dead through the brilliance of the Father, we also may carry on with another life" (See Romans 6:4, the foregoing is a paraphrase).
