The phrase "the spirit who now works in the sons of disobedience" is a further description of the being referred to in the preceding phrase in the verse ("the prince of the power of the air").
"The prince of the power of the air" is a title of Satan, and is based on the realm of activity that Satan possesses in this fallen world, and the medium in which he and his demons move and operate. He is given the title "prince" because the permissive will of God in this present age still allows him to exercise this authority (1 John 5:19). But he is ultimately under the control of the King (that is, Christ), and will be eternally confined and tormented at the close of the age, as described in Revelation 20:7-10.
